Circulating tumor cells and DNA in early diagnosis and prognosis of metastatic cancer
Abstract
Metastatic cancer remains a formidable challenge in oncology and frequently presents with limited intervention options at late-stage diagnosis. This study highlights the role of circulating tumor cells (CTCs) and circulating tumor DNA (ctDNA) as biomarkers for early detection and diagnosis and prognosis. Liquid biopsy analyzes blood components like CTCs which provide insights into tumor heterogeneity and metastatic potential and ctDNA which reflects genetic mutations and tumor burden. We elucidated the biological properties of CTCs which are viable intact cells that detach from the primary tumor and enter the bloodstream whereas ctDNA consists of small fragments of DNA released through apoptosis and necrosis. The analysis highlights advances in detection methodologies such as Next-Generation Sequencing and droplet digital PCR which facilitate real-time tumor monitoring and the assessment of minimal residual disease. The comparative assessment revealed that CTCs facilitate transcriptome and proteome profiling whereas ctDNA allows for the detection of specific mutations and copy number variations. The presence of these biomarkers correlates with disease progression and treatment resistance and offers predictive value for patient outcomes. The integration of these biomarkers into clinical practice holds the potential to revolutionize metastatic cancer management through individualized strategies and improves early detection outcomes. This comprehensive review underscores the necessity of standardizing detection protocols to overcome biological and technical challenges and ensures the successful clinical implementation of liquid biopsies in precision oncology.
Biography
Marium Jahan is currently pursuing a PhD in the Department of Biochemistry at Quaid-i-Azam University, Pakistan. Research activities are being conducted under the supervision of Prof. Dr. Aneesa Sultan in the Cancer Genetics Laboratory, with specialization in cancer research, genetics, and molecular biology. Current research is focused on breast cancer, with particular emphasis on investigating the role of circulating tumor cells and genetic biomarkers in metastatic disease progression. Extensive technical expertise has been developed in molecular biology techniques, including PCR, DNA sequencing, gene regulation analysis, and gel electrophoresis. Previous contributions have also been made to the study of urologic cancers and gastrointestinal stromal tumors. Through this research, efforts are directed toward elucidating the molecular mechanisms underlying cancer progression, facilitating the development of personalized treatment strategies, and addressing critical challenges associated with early diagnosis and prognosis.
